You probably want to see some wildlife. Lamar Valley, the Tower/Roosevelt area, Mt. Washburn and then Hayden Valley are some of the better places to see wildlife, but you can encounter just about anything, anywhere in the park.
You'll want to spend some time in the geyser basins. Old Faithful, of course, but do visit 3 or 4 of the others as well.
The canyon is a do not miss as well. If you are up to it, hike down to the brink of the Lower Falls. It's about a 400 feet climb back up though, so keep that in mind. Very much worth the effort if you are able.
